Non-Standard DNA Samples: Are They as Accurate as Buccal Swabs or Blood Tests?

When most people think about DNA testing, they picture a simple cheek swab or, less commonly, a blood sample. While these remain the gold standard for DNA collection, there are situations where obtaining a standard sample isn't possible. In these cases, forensic laboratories may be able to extract DNA from what are known as non-standard or alternative DNA samples.

But how reliable are these samples? Can they produce the same results as a cheek swab? Let's explore.

What Is a Standard DNA Sample?

A standard DNA sample is one specifically collected for DNA testing under controlled conditions. The two most common types are:

  • Buccal (cheek) swabs – The preferred method for most paternity, maternity, and relationship tests.

  • Blood samples – Once the standard for DNA testing but now rarely necessary because cheek swabs provide the same DNA profile without the discomfort of a blood draw.

Both sample types contain thousands of intact human cells, allowing laboratories to produce high-quality DNA profiles with an extremely high success rate.

What Are Non-Standard DNA Samples?

Non-standard DNA samples are everyday items or biological materials that may contain enough human DNA for testing. These samples are often used when a person is unavailable, deceased, or unwilling to provide a cheek swab.

Examples include:

  • Hair with the root attached

  • Toothbrushes

  • Electric razor cartridges

  • Used razors

  • Fingernail clippings

  • Nail scrapings

  • Cigarette butts

  • Chewing gum

  • Used tissues

  • Drinking straws

  • Drinking cups or bottles

  • Dental retainers

  • Dentures

  • Earbuds (Q-tips)

  • Lipstick

  • Baby pacifiers

  • Medical specimens

  • Bone or teeth (for deceased individuals)

Not every item listed will contain usable DNA. Success depends on how much human DNA is present and whether the sample has been contaminated or degraded.

Are Non-Standard Samples as Accurate?

This is one of the most common questions we receive.

The answer is yes—with an important qualification.

If the laboratory is able to obtain a complete DNA profile from a non-standard sample, the comparison is just as accurate as one performed using a cheek swab or blood sample.

The DNA itself does not change based on where it came from. Whether DNA is extracted from a cheek swab, blood, toothbrush, or hair root, the genetic profile belongs to the individual and produces the same scientific comparison.

The challenge is obtaining enough high-quality DNA.

Why Buccal Swabs Are Still Preferred

Although non-standard samples can work well, cheek swabs remain the preferred collection method because they offer several advantages:

  • Highest chance of obtaining a complete DNA profile

  • Fast laboratory processing

  • Minimal risk of contamination

  • Simple, painless collection

  • Lower testing costs

  • Reliable chain of custody for legal testing

For these reasons, accredited laboratories always recommend a cheek swab whenever it is available.

Challenges with Alternative Samples

Unlike a fresh cheek swab, non-standard samples may present several challenges:

Limited DNA

Some items simply do not contain enough human cells for testing.

DNA Degradation

Heat, sunlight, moisture, and age can damage DNA over time.

Mixed DNA

Objects handled by multiple people may contain DNA from more than one individual, making interpretation difficult.

Contamination

Improper handling can introduce DNA from another person, potentially affecting the testing process.

Because of these factors, laboratories cannot guarantee that every non-standard sample will produce a usable DNA profile.

Which Non-Standard Samples Work Best?

Generally, laboratories have the highest success rates with:

  1. Toothbrushes used regularly by only one person

  2. Hair with the root attached

  3. Used razors

  4. Fingernail clippings

  5. Baby pacifiers

  6. Fresh cigarette butts used by only one individual

Items such as cut hair without the root, old tissues, or objects handled by multiple people are less reliable.

Can Non-Standard Samples Be Used for Legal DNA Testing?

In most cases, no.

Legal DNA tests require identity verification and a documented chain of custody. Because the origin of a non-standard sample usually cannot be independently verified, these samples are generally suitable only for informational testing.

If you need DNA results for:

  • Immigration

  • Court proceedings

  • Child support

  • Birth certificate amendments

  • Inheritance matters

a properly collected legal cheek swab is almost always required.

How Should Alternative Samples Be Stored?

If you plan to submit a non-standard sample:

  • Handle it as little as possible.

  • Wear disposable gloves if available.

  • Allow wet items to air dry completely.

  • Place each item in a separate paper envelope (avoid plastic bags when possible).

  • Label each sample clearly.

  • Keep samples in a cool, dry place until they are shipped.

Proper storage helps preserve DNA and improves the likelihood of successful testing.
